Abstract

For the last several decades, X-ray crystallography has been the cornerstone of protein structure determination. Such structure determination have been great enablers in understanding how key protein-protein and protein drug interactions happen at a molecular level. Despite this, the structure determined by X- ray crystallography offers a static snapshot of the protein/macromolecule in the crystal. In solution, a protein can potentially sample multiple conformation(s) that may be important for mediating its function. Such a dynamic view of protein structure can be visualized by NMR spectroscopy. In this talk, the role of protein dynamics in enzyme activity shall be illustrated in the case of L-asparaganise, a protein drug that is used for treatment of childhood leukaemia. Extension of the methodology, developed in the lab, in case of therapeutic monoclonal antibody shall be touched upon.
